Abstract

Cibotium barometz (L.) J.Sm. (Cibotiaceae) is an important export commodity for both traditional and mod-ern medicine. Populations of this species in several countries have decreased rapidly due to the uncontrolled collection of the rhizome parts for medicinal purposes. Since 1976, this species has been included in Appendix II of the Convention on International Trade in Endangered Species (CITES). This means that no export is allowed without a prior permit issued by the CITES committee.  In order to utilize an endangered species sustainably, the global NDF (Non-Detriments Finding) system is applied for determining annual quotas. Therefore, monitoring and updating the inventory of C. barometz in its natural habitat should be carried out annually. A population study of C. barometz carried out in 2011 in Riau Province, Sumatra, is reported here. The aims of the study were: 1) to inventory C. barometz and determine its variation in Riau Province, Suma-tra, 2) to study the distribution and ecology of C. barometz, and 3) to assess the population size of this spe-cies by using random search methodology incorporating belt line transects. Two variants of C. barometz are recognized; they are the golden yellow and golden brown variants. C. barometz is distributed in eight loca-tions of Kampar District of Riau Province, in the secondary forest and rubber agroforest between 80 m and 600 m above sea level (asl). This species grows well in open to partially opened areas of secondary forest and rubber plantation in hills with a range of slope between 30° and 90°, with a relatively high humidity, 60 – 90%, in acid to nearly neutral soil, with a range of soil fertility from very poor to very humus rich soil.  The average population density determined in our study was 20 plants per 100 square meter. The highest popula-tion size was in the secondary forest of Bukit Kuda Beban at 590 – 600 m asl., viz. 9405 plants with a popula-tion density  of 47 plants per 100 square meter.

Abstract

Indonesia is a high-abundance tropical forests country. It plays a very important role for world life because of its species richness than others. One of the forest resources in Indonesia is indigenous forests. Indonesia is rich of local wisdom such as that possessed by indigenous peoples. Indigenous peoples have a potential of the biological resources conservation. As a sustainable management tradition, communities around indigenous forest also have the efforts to preserve the forest.  The data collection of community knowledge about plants was conducted by interviews and direct-field observation. Data were analyzed using index of cultural significance. The results showed that there are 48 species of plants utilized by communities in Hiang Indigenous Forest, 27 species are used as firewoods, 15 species as building materials, 7 species as medicinal plants, 6 species as traditional handicraft ingredients and 4 species as secondary foods and traditional ritual materials. Most of widely-used plants by the community are Altingia excels, and Styrax benzoin is used as incense in a traditional ritual.

Abstract

Upregulation of nephrin expression occurs at the early stage of nephropathy and decrease in the period 6 months on hyperglycaemic condition. Nephrin is associated with the initial stage of the loss of the permeability barrier in diabetic nephropathy. Interaction AGE-RAGE increases angiotensin II on Renin Angiotensin-Aldosterone System (RAAS) and activation of protein kinase c (PKC) which induce alterations in nephrin mRNA expression. Alterations of nephrin expression induces transformation of slit membrane structure and the permeability changes at the glomerular filtration barrier. Anti-AGE vaccination once may cause the changes of nephrin and RAGE expression and can prevent progression of diabetic nephropathy. This study used primary glomerulus cell culture obtained from renal of Wistar mice aged 3 months, weighting 200-300 grams that consist of negative control group that exposed to BSA 100 µg/ml, positive control group that exposed to AGE-BSA 100 µg/ml, treatment group 1 that exposed to polyclonal anti-AGE 5 µg/ml and AGE-BSA 100 µg/ml and treatment group 2 that exposed to monoclonal antibody anti-CML 5 µg/ml and AGE-BSA 100 µg/ml. Paired t-test with a 0.05 level of confidence results showed that there were significant differences in level of RAGE expression among experimental groups with control groups. Administration of polyclonal antibody decreased RAGE expression among negative control (p=0.188). but not in positive control (p=0.000). In contrast to monoclonal anti-AGE antibody, RAGE expression did not differ significantly compared to negative control but significant than positive control. Administration of monoclonal anti-AGE antibody inhibited increasing of nephrin expression compared to negative and positive control (p=0.73; 0.125). In conclusion, this study suggested that administration of polyclonal and monoclonal anti-AGE antibody could inhibit increasing of RAGE and nephrin expression in glomerulus primary culture that exposed to AGE which is expected to prevent the progression of diabetic nephropaty.

Abstract

Rheumatic heart disease (RHD) is mediated by an abnormal immunological response following a Streptococcus pyogenes infection that induces a disturbance of oxidants and antioxidants balances. Mannose-binding lectin (MBL) binds to N-acetylglucosamine, a molecule present on the Streptococcus cell wall and human heart valves. There is a disturbance of oxidant and antioxidant balance in rheumatic disease. Myeloperoxidase (MPO) is a marker of oxidative stress and inflammation. This study was aimed to determine the correlation of MBL and MPO levels and severity of valvular regurgitation and heart failure (HF) in RHD patients. A case-control study was conduct using human peripheral blood samples from 32 children aged 6 to 14 years old. The subjects were divided into two groups: 16 RHD patients included in the case group and 16 healthy children as a control group. The level of MBL and MPO was investigated using ELISA method. There were significant differences on MBL and MPO level between patient and control group. The level of MBL and MPO were significantly increased in RHD group, especially on severe valvular regurgitation. There was a strong correlation between MBL and MPO levels and the severity of valvular regurgitation (r = 0.94 and r = 0.88). The least significant diff-erence (LSD) analysis showed that significant difference occurs in the severe heart failure group. Our research revealed that the MBL and MPO levels in pediatric RHD patients were significantly higher than in healthy children. The MBL and MPO levels were significantly correlated with the severity of valvular regurgitation and heart failure.

Abstract

Nusa Tenggara consists of some small islands, one of them is Moyo Island. The diversity of plant species in this island is not really known for certain. This research was determined to observe the diversity of plant spe-cies in Moyo Island forest. The research was conducted in April 2013 by using floristic analysis method. The characteristic of Moyo Island forest is lowland evergreen rain forest. The results showed there were 60 tree species recorded in Moyo Island forest. There were many fruiting trees and seedling from the trees which show healthy growth, indicated that the plant regeneration in this forest is went well. The diversity of Pteri-dophytes and orchids were not high. Epiphytic fern which often found in the forest were Drynaria quersifolia and Platycerium bifurcatum and terrestrial orchid that dominated in the forest was Nervilia aragoana. Tuber plant was often found in this forest and grew prolifically were Tacca, Dioscorea and Amorphophallus. In coastal area lived a population of Pandanus tectorius. There were three new record plants found. The first was epiphytic orchid: Pteroceras javanica, the second was the epiphytic plant: Hoya verticillata and wild tuber plant: Tacca leontopetaloides.

Abstract

Aedes aegypti is the vector of Dengue Hemorrhagic Fever (DHF) disease. Containers are breeding places for DHF vector and the most commonly found larvae in the bath water containers. The presence of Aedes aegypti larvae in a container could be affected by the type of water source besides the container’s color, material, location, lid existence and the container’s drain frequency. This study was conducted to determine the associate of water source type with larvae presence and the additional factors. This study used observational analytic with case control design. The case group consisted of households using well water and the control group consisted of households using tap water with a sample size of 130 households for each group. The sample was collected by proportional random sampling in five villages. The data was analyzed using a regression logistic test. The significant variables associated with the presence of larvae were the water source type (OR=1.923), container’s color (OR=2.345), container’s location (OR=2.241), container’s lid existence (OR=2.122) and the container’s drain frequency (OR=2.260). This study did not consider the significant association of the container’s material. The dominant variable associated with the presence of larvae was the container’s drain frequency which was controlled by the water source type, container’s color and container’s location.

Abstract

P-glycoprotein (P-gp) resistance in cancer cells decreases intracellular accumulation of various anticancer drugs. This multidrug resistance (MDR) protein can be modulated by a number of non-cytotoxic drugs. We have screened 30 chincona alkaloids derivatives as a potent P-gp inhibitor agent in silico. Hereby, we report the highest potential inhibitions of P-gp is Cinchonidine isobutanoate through molecular docking approach. with affinity energy -8.6 kcal/mol and inhibition constant, Ki is 4.89 x 10-7 M. Cinchonidine isobutanoate is also known has molecular weight below 500, Log P value 3.5, which is indicated violation free of Lipinski`s rule of five. Thus, Cinchonidine isobutanoate is the most potent compound as anticancer compare to other Cinchona alkaloids. Ultimately, we design Cinchonidine isobutanoate for further lead synthesis by using DBSA, act as a combined Brønsted acid-surfactant-catalyst (BASC) to obtain high concentration of organic product by forming micellar aggregates which is very powerful catalytic application in water environment.

Abstract

Breast cancer (BC) is the most common type of cancer worldwide, being a major cause of morbidity in female. In Nepal it is the second most common type of cancer among females of perimenopausal age group. More than one quarter of the BC diagnosed in young Nepalese female, with familial history of breast cancer, early pregnancy, longer lactation and estrogen exposure and often with tumors showing aggressive biological behaviors. Anthracyclines (Doxorubicin) based treatment regime were reported to cause cardiotoxicity by increasing intramyocardial free radical production, lipid alterations and decreasing antioxidant level. Oxidative stress involving cellular reactive oxygen species (ROS) production is widely accepted mechanism but the molecular basis of chemotherapy induced organ toxicity remains highly controversial. An increased rate of metabolism and oxidative stress results rapid turnover of cancer cells that modulates the enzyme level in blood circulation. Serum LDH and GGT level correlates with tumor burden, metastatic character of BC and intensity of organtoxicity. The aim of our study is to evaluate the serum level of LDH and GGT in BC patients receiving chemotherapy and correlate these enzyme levels with different stages of BC. A total number of 150 subjects were included in the study, comprising 90 histopathologically confirmed 24 to 76 years aged patients of different breast cancer stages, receiving at least 3 cycles of 5-Fluorouracil, Adriamycin and Cyclophosphamide (FAC) chemotherapy. Sixty age- matched healthy women were enrolled as controls. Blood samples from each individual was collected after informed consent and analysed for serum LDH and GGT levels using standard biochemical methods. Data were analyzed using student's paired 't' test, Pearson correlation test and ANOVA. Serum LDH and GGT levels were significantly (p<0.001) increased in BC patients as compared to control group. When all 4 stages of BC were compared to control group, LDH and GGT showed steady and progressive increase in the activities of these enzymes from stage I-IV. The study concludes that serum LDH and GGT may be sensitive, specific and cost effective biomarkers in early diagnosis of breast cancer, assessing cancer prognosis and response to treatment.

Abstract

Dengue control efforts have not provided adequate results. Therefore we need other measures like sterile in-sect Technique (SIT). This study aimed to observe the level of egg sterility and age of Aedes aegypti mosqui-toes after using SIT in Ngaliyan Semarang. The method of this research is a descriptive study with a cross-sectional design. A sample of the mosquitoes eggs are all the mosquitoes eggs on the paper trap in every house of RW2 population in Ngaliyan village. These samples are the result of the fertilization of male and female mosquitoes in nature. Data analysis using Wilcoxon sign test. The result showed that the average of egg sterility percentage in mosquitoes after using SIT indoors was 73.65% and the average of egg sterility per-centage outdoors was 69.08%. Wilcoxon test showed significant differences in average age of mosquitoes egg (p < 0.05), males pupae (p < 0.05), females pupae (p < 0.05), and adult female mosquitoes (p < 0.05), before and after SIT. It can conclude from this research that the SIT release in Ngaliyan village Semarang city could increase the level of sterility mosquito’s eggs and a decrease of the age of Ae. aegypti mosquitoes in this area.

Abstract

The cyclic structure of ADTC5 (Ac-CDTPPVC-NH2) peptide is known has ability to modulate homo dimer E-cadherin interactions to form adherens junction at the intercellular junction.  The ability to inhibit E-cadherin interaction has become important to increase paracellular porosity in delivering drug molecules to the target cell. There are two types of ADTC5 state: opened-cyclic state (OCS) and closed-cyclic state (CCS). OCS of ADTC5 is affected by distance constraint and CCS is formed by disulfide bond from terminal cysteines through force restraint. The aim of this research is to investigate the inhibition activity of ADTC5 peptide upon E-cadherin. Here we used molecular docking and molecular dynamics approaches. The structure of ADTC5 peptide was generated by PyMOL program. GROMACS v4.5.5 was utilized to simulate molecular dynamics. The optimized ADTC5 molecule was placed in aqueous or polar condition at physiological pH. Furthermore, ADTC5 was docked with EC1-EC2 coupled domain of E-cadherin using AutoDock 4.2 and be refined using molecular dynamic (MD). The result showed that CCS ADTC5 peptide has stronger affinity and more stable interaction with EC1-EC2 coupled domain than the OCS one.
